Abstract

Background: A2B adenosine receptor (A2BAR) is member of the family of seven transmembrane G protein coupled receptor that are activated in high concentration of extracellular adenosine that occur in certain pathophysiological conditions. Activation of the A2B adenosine receptor in tumor cell line limits or promotes tumor growth. In current study, we investigated the role of A2B adenosine receptor-agonist in apoptosis induction in ACHN renal cancer cell line. Methods: ACHN renal cancer cell line was selected to investigate the effect of A2B adenosine receptor-agonist (BAY 60-6583) on apoptosis. The cells were cultured in high glucose Dulbecco's Modified Eagle's Medium (DMEM) (10 FBS and 1 penicillin/streptomycin). Cell viability and apoptosis were evaluated using 3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium bromide (MTT) assay and Annexin V/propidium iodide (PI), respectively. Findings: Based on MTT assay, treated samples with 50, 100 and 200 μmol of BAY 60-6583 had approximately 50 viability after 48 hours. In addition, based on Annexin V/propidium iodide test, 50 and 100 μmol concentration of BAY 60-658 induced the most apoptosis after 48 hours. Conclusion: According to the results, it can be concluded that BAY 60-6583 induces apoptosis in ACHN cell line with A2B adenosine receptor stimulation, therefore the A2B adenosine receptor-agonists can be used as agent that inhibit cancer growth. © 2018, Isfahan University of Medical Sciences(IUMS).
